The PathHunter eXpress U2OS JAK3 Activity Assay is a stable, engineered cell line for use in studying small molecule drug candidates targeting the JAK3 kinase domain. The cell line is engineered to express the catalytic domain from JAK3 fused to the cytokine receptor binding domain (CRBD) of JAK2. The cells co-express an Enzyme Acceptor (EA) tagged SH2 domain and a ProLink (PK) tagged Cytokine receptor. The CRBD is designed to bind the CTK catalytic domain to the PRLR-PK. Activation of the PRLR-PK induces receptor phosphorylation by the JAK3 catalytic domain. In the floating matrix contraction model, a freshly polymerized collagen matrix containing cells is released from the culture dish and allowed to float in culture medium, and contraction occurs in the absence of external mechanical load and without appearance of stress fibers in the cells. The CytoSelect™ 48-Well Cell Contraction Assay Kit provides a simple system to assess cell contractivity in vitro and screen cell contraction mediators using the floating matrix model. The kit's proprietary Cell Contraction Plate eliminates the matrix releasing step of the conventional contraction assay, providing a faster, higher-throughput method to assess cell contraction.
